3 EXPERIMENT NO:2 DATE - 11/10/22 ISOLATION OF BACTERIA FROM WATER AND AIR SAMPLE AIM to whats Instruction fean environmental sample MATERIALS REQUIRED 1 Nutrient agae 2. Steele swates 3 Felli plates 4. Test tubes PROCEDURE FOR ISOLATION OF BACTERIA FROM AIR 1) 150 ml of nutrient agne was prepared as placed in the auto lave along with are petrip lates for stecilization 2) After stecilization ,the nutrient agar was pareed in the petriplates al allowed it to solidity 3) After the plates all solidified, all the plates all taken air all the laminar air flow of kept in the open for different of internals of lime ie, 5min (s splates) 10min (2 plates) El 15 min (2 plates). 4) / plate is kept sepalately inside Laminae All How for 5) linted Ague exposure of plates to air , it is incubated at 37°C for 24 hours 6) After 24 houes the bacterial wonces were observed. OBSERVATION The bacterial colonies were formed at different time internal and different merphology of colonies well obseeved. RESULT 12 whonics well formed when the plates well exposed for 5 min , 24 colonics well formed when the plates well exposed for 10min , 30 colonies well formed on the plates when exposed for IS mins.
4 PROCEDURE FOR ISOLATION OF BACTERIA FROM WATER 1 150 ml of nutritional agai was prepared 2. 7 tubes containing 9ml of each dissilled water was prepared 3 Test tubes peticiplates up nutrement agal was kept for steriligation using autoclave 4 type stecilization add Ind of water sample to the test tube containing Iml of distilled water (This is 10 -1 dilution) 5. FROM each dilution 1ml was taken as made upto 10 dilution 6 Nutrient agar was powed in the petriplates 4 allowed it to solidity ? After serial dilution af solidification of nutrient agal 1ml of sample was pepetted into the petriplates The 0 entire surface was carefully spreaded using steele L shaped 8 glass rod. 9. The plates well incubated at 37°C 101 24 haves. 10. After 24 have of incubation the bacterial colonies were observed al calculated RESULTS Bacterial donies with different characteristics were found to be grown in both pend and tap water DISCUSSION Unlike soil 4 water ail does not contain nutrient material to support growth of micro organisms it acts as medium for transfer of mirobial propagules Microbes in air generally come from sal water al living organisms activities. The micropora of air is highly dynamic af is affected by timp, wind, humidity 4 other human El animal activities life on earth It may become the place for the growth of water form a major source is supporting all forms differe types of of microoyanisms which may have a potential for spreading a variety of diseases. The mal the amount of bacteria in water the more it is to drink Regular isolation of bioarcesds 4 water nicobes helps to understand the quality of air 4 water.
