7 EXPERIMENT NO:4 DATE 11/10/22 HANGING DROP METHOD AIM To find out the motility of bacteria MATERIALS REQUIRED CAVITY GLASS SLIDE, coverslip / petroleum jelly, bacterial culture , microscope , nicheone will inouranting loop PROCEDURE / like clear cavily slide at coverslip. 2. Apply vascline / glue around the cavely on the slide Ef wences of coverslip 3 Place a drop of bacterial velture in the center of coverslip 4 carefully keep the cavity of the slide over the welture on coverslip al genlty press the slide al now the bacterial withill hangs. Reverse the slide , 80 that coverslip is on top of the cavity 5 6 observe just in 10X El later 40x. OBSERVATION Many rod shaped 4 circular shaped bacteria well observed moving in the slide. RESULT True motility of rod shaped bacillus El spherical coll were found under the mivoscope . They change position with respect to one al another DISCUSSION Hanging deop preparation is a modified version of the wet mount method - Hell a drop of medium containing the organisms is plated on a microscopic slide , open used in dark illumination to observe the motility of bacteria Motility is the movement ability of bacteria from one direction to another by their our same bacteria all motile al some all non - motile. The self propulsion
