20 EXPERIMENT - 8 DATE - 15/11/22 CARBOHYDRATE FERMENTATION TEST AIM To determine the ability 7 the given microganisms to toement the (aebchydealt as production of organic and and product of gasious end product MATERIALS REQUIRED Testtubes Jest tube steand, carbohydrate l alucose mannitol success , basal medium, Duesham's tube, Phaned red (indicator) PROCEDURE 1) we weighed the oxidation termentation basal medium for 120 ml souli on El transfered into conical Hasks 2) Distrilled water added to prepare a solution. 3) After preparing it is-kensferred to 4 different conical Masks, to the discred carbohydeate can be added 4) We added 1g of glucose , sucrose , mannitol al galaclose into the 4 different conical flask 5) Phend red was added as an indicator 6) 30 ml solution of each onical flask is distributed in tubes each containing 10ml. (2 tubes fa glucose 3 tubes for success, 3 tubes for mannild 3 tubes for galancose) 7) Then we insealed the Dusham's tube in a invest position in all the tubes 8) All the tubes were kept far starilization 9) After sterilization 4/ cooling of the tubes organisms were inaulated into 8 wbes keeping 4 tubes as conted with the help of inoulation loop 10) After ingulation the tubes are kept for immunition for 24 hes at 37°C
21 11) Aller 24 hes of incubation the tubes all examined for acid 41 gas product OBSERVATION the tukes in which glucose was present as the caebo hydrate the colour of tube changed to yellow but the durshen tube was present at the bottom A The tubes in which sucrose was present as the carbohydees there was no whour change it the duesham hube was at the bottom the tubes in which mannitol was present as the carbohydea , there was whoul change al dusham tube was Heating The tubes in which galactose was present as the carbohydeal / there was no colour change Ell the durham tube was present at the bottom . RESULT From the above observation the given organism is able to ferment glucose by producing acid but thell is mo gas production. The organism is able to ferment mannital by producing acid as well as gas but the organism is not able to terment sucrose of galactose as the carbohydrate. DISCUSSION Bacteria can be differentialed based on their abeling to respire (aerobically ,anaeoobically or facultative) or their ability to produce ATP via termentation The carbohydrate termentation lest is used to detect whether or not a bacteria can utilize ,a certain carbohydeate . It lests for the presence of acid al/os gas produced from the termentation of a single particular carbohydrate.
